Glucocorticoids suppress macrophage migration inhibitory factor (MIF) expression in a cell-type specific manner

Alourfi Z , Donn R , Stevens A , Ray D

Aim: To investigate glucocorticoid (Gc) regulation of MIFBackground: MIF is a potent proinflammatory cytokine involved in inflammatory arthritis. Previous work suggested that MIF was induced by low concentrations of Gc, and may counteract their anti-inflammatory effect. The relationship between MIF and Gc has not been investigated in human cells. Methods: The human cell lines CEMC7A (T-lymphoblast) and A549 (lung epithelial) were us...

Proinflammatory cytokine gene expression shows intrinsic circadian oscillation in peripheral cells

McMaster A , Lebiecki J , Loudon A , Donn R , Ray DW

An extraordinary array of behavioural and physiological systems is regulated by endogenous circadian timekeepers within mammals. This circadian machinery is comprised of intricate autoregulatory transcription feedback loops, which interact to generate a time delay of approximately 24 hours in the cyclical transcription of components. In addition to the master clock in the suprachiasmatic nucleus it is clear that peripheral tissues also express clock components. The central clo...

Modulation of cortisol action

Jangani M. , Berry A. , Matthews L. , Farrow S. , Donn R. , Ray D.

Variation in glucocorticoid (Gc) sensitivity underlies metabolic disease, and affects therapeutic response in inflammation. Using an in-vitro screening approach we identify two new mechanisms capable of regulating Gc sensitivity. The first involves interferon induced factor 16 (IFI16). IFI16 potentiated both transactivation, and Gc repression of NFκB. IFI16 did not affect glucocorticoid receptor (GR) expression, ligand dependent repression of GR expr...
